چکیده

PPAR and C/EBP are critical transcription factors in adipogenesis, but the precise role of these proteins has been difficult to ascertain because they positively regulate each other’s expression. Questions remain about whether these factors operate independently in separate, parallel pathways of differentiation, or whether a single pathway exists. PPAR can promote adipogenesis in C/EBP -deficient cells, but the converse has not been tested. We have created an immortalized line of fibroblasts lacking PPAR , which we use to show that C/EBP has no ability to promote adipogenesis in the absence of PPAR . These results indicate that C/EBP and PPAR participate in a single pathway of fat cell development with PPAR being the proximal effector of adipogenesis.

Inhibitory Effects of Hwangryunhaedok-Tang in 3T3-L1 Adipogenesis by Regulation of Raf/MEK1/ERK1/2 Pathway and PDK1/Akt Phosphorylation

Hwangryunhaedok-tang (HRT) has been long used as traditional medicine in Asia. However, inhibitory role of HRT is unclear in early stage of 3T3-L1 adipocyte differentiation related to signaling. In the present study, we investigated the inhibitory effects of HRT on upstream signaling of peroxisome proliferation-activity receptor- γ (PPAR- γ ) and CCAAT/enhancer binding protein- β (C/EBP- β ) ex...

Sodium arsenite inhibits and reverses expression of adipogenic and fat cell-specific genes during in vitro adipogenesis.

Arsenic causes cancer in humans, but its mechanism of action is unique among known carcinogenic agents. As a naturally occurring component of sediments and ground water, human exposure to arsenic is inevitable, necessitating the establishment of exposure limits. Because cancer is characterized as an imbalance between cell growth and differentiation, it has been hypothesized that arsenic exerts ...

Phosphorylation of C/EBP at a Consensus Extracellular Signal-Regulated Kinase/Glycogen Synthase Kinase 3 Site Is Required for the Induction of Adiponectin Gene Expression during the Differentiation of Mouse Fibroblasts into Adipocytes

Stimulation of adipogenesis in mouse preadipocytes requires C/EBP as well as activation of the MEK/ extracellular signal-regulated kinase (ERK) signaling pathway. In this study, we demonstrate that phosphorylation of C/EBP at a consensus ERK/glycogen synthase kinase 3 (GSK3) site regulates adiponectin gene expression during the C/EBP -facilitated differentiation of mouse fibroblasts into adipoc...

Sequential phosphorylation of CCAAT enhancer-binding protein by MAPK and glycogen synthase kinase 3 is required for adipogenesis

CCAAT enhancer-binding protein (C EBP) , C EBP , and peroxisome proliferator activated receptor (PPAR) act in a cascade where C EBP activates expression of C EBP and PPAR , which then function as pleiotropic activators of genes that produce the adipocyte phenotype. When growth-arrested 3T3-L1 preadipocytes are induced to differentiate, C EBP is rapidly expressed but still lacks DNA-binding acti...

Transdifferentiation of myoblasts by the adipogenic transcription factors PPAR gamma and C/EBP alpha.

Skeletal muscle and adipose tissue development often has a reciprocal relationship in vivo, particularly in myodystrophic states. We have investigated whether determined myoblasts with no inherent adipogenic potential can be induced to transdifferentiate into mature adipocytes by the ectopic expression of two adipogenic transcription factors, PPAR gamma and C/EBP alpha. When cultured under opti...
